Concentration of cryo-solution versus time for (a) a sequential 1 min soak in each of 10, 20, 30 and 40% solutions, transferring the crystal between solutions, and (b) leaving the crystal in the same place in a 10 µl drop of mother liquor and pipetting 10 µl of 10% cryo-solution onto it, mixing, waiting 30 s, removing 10 µl, adding another 10 µl of 10%, mixing, waiting 30 s, removing 10 µl, adding 10 µl of 20% and so on. In (b) the crystal is subjected to more gradual osmotic shock as the concentration of cryo-solution is increased.
