Data-collection geometry for 2D crystals. Each lattice line will have a continuously varying amplitude and phase along its Z coordinate. The tilted views sample the lattice lines at different heights in Fourier space. Most of the 3D FT can be determined by combining these sections derived from different tilt views in projection. However, in EM of 2D crystals there is a missing cone of information because the maximum tilt angle of the crystal is limited to about 60°. Figure adapted from Amos et al. (1982  ). |
