Helical diffraction. A helix can be considered as a 1D crystal, since it has a repeating structure along the axis giving rise to a set of layer lines in the diffraction pattern. If the symmetry of the helix is known, a full 3D reconstruction can be calculated from the untilted filament transform, since the subunit is imaged at different angles about the filament axis. The example shown is for the case of a helix with an integral number of subunits per helical pitch repeat. |
