Cryo-electron microscopy of Semliki Forest virus. Selected regions from different cryo-electron micrographs of SFV particles which were used for image processing and three-dimensional reconstruction. The effect of defocusing on the appearance of viral features is shown for δf = 1 (a), 2 (b), 3 (c) and 4 µm (d). The theoretical phase contrast transfer functions for these defocus values are shown in the plot in (e). Note that further defocus values would be necessary to give complete coverage to 7 Å resolution. |
