Influence of culture conditions on soluble protein production: OD₆₀₀. Plasmids were transformed in RosettaII (DE3) strain and induced during early (E), mid (M) or late (L) logarithmic growth phase or during early stationary phase (S) using 1 mM IPTG for 14 h at 293 K. Extracts prepared with PopCulture were affinity-purified using MagnaHis beads and analyzed by SDS–PAGE. Examples are shown where the OD₆₀₀ value influences soluble expression.
