Kd values were determined by nonlinear regression using a single-site model as indicated by the linearity of the Scatchard plot (inset). (b) Determination of the ratio of TPP+ binding to EmrE. The concentration of EmrE in the experiment was determined by amino-acid analysis to be 80.5 µM and the intersection between the linear portions of the graph occurs at a TPP+ concentration of 40 µM.
