One heterodimer is colored cyan (AscX31) and magenta (YscY), while the other is shown in gray (AscX31) and black (YscY). The inset highlights the covalent linkage between Cys23 of the two YscY chains in the asymmetric unit. The 2mFoDFc density is contoured at the 1σ level. (b) Analytical size-exclusion chromatography using a Superdex 75 10/300 GL column (Cytiva) produced a single peak for AscX31–YscY at the expected weight for a 1:1 complex. No cross-linked species were observed on a polyacrylamide gel.
