Correlation of synchrotron and cryo-EM imaging of frozen-hydrated apoferritin. (a) Apoferritin was plunge-frozen in liquid ethane. (b) The grid was clipped into AutoGrid rings and mounted onto custom 3D-printed holders. (c) At the beamline, the sample was transferred into the loading carrier, optically monitored with a camera and exposed to varying absorbed X-ray doses across selected grid squares. (d) The same grid squares were subsequently imaged by cryo-TEM and data were recorded. |
