Apoferritin single-particle data analysis and final resolution estimation. (a) Complete electron-density map of apoferritin from 6488 particles without X-ray exposure. (b) The Fourier shell correlation analysis of the three maps yielded resolutions of 3.17 Å for the dataset without X-ray exposure and 3.56 and 3.88 Å for the apoferritin samples which had absorbed 1 and 100 MGy X-ray doses, respectively. All three reconstructions were based on the same number of particles. These results indicate that higher X-ray exposure doses caused progressive protein damage, leading to a loss of resolution.
