Differential deuteriation in the thermodynamic cycle. The determined binding Kd values (mM) of melibiose or Na+ in the absence or presence of the other are presented in the binding thermodynamic cycle scheme, adapted from Fig. 6 of Hariharan & Guan (2017View full citation). The differential deuteriation was calculated from the deuterium uptake (D) at 30, 300 and 3000 s in two states as indicated. All data (>threshold and P < 0.05) were mapped to the outward-facing structure and displayed in two orientations related by 180°, as indicated. N- and C-termini are labeled, and membrane orientation is indicated. Blue-colored regions, protection; red-colored regions, deprotection; purple-colored region, deprotection at 30 s and protection at 3000 s.
