The loading and activation of MCM rings onto DNA for replication involves transformations between open and closed rings because MCM rings are placed at interior regions of the DNA and are not threaded like a needle onto the ends. (a) During loading, MCM open rings are loaded onto DNA to generate two closed rings encircling DNA as a double hexamer (see PDB entries 5v8f and 5bk4; Yuan et al., 2017; Noguchi et al., 2017). (b) Activation of the MCM ring to unwind DNA involves the sequential recruitment of Cdc45, GINS and Mcm10 (Douglas et al., 2018). The closed rings of the double hexamer must open to remove one of the strands from the ring pore. |
