Preparation of frozen-hydrated biological samples. (a) Procedures for pre-treatment of the sample support disc and preparation of frozen-hydrated biological samples under controlled RH. (b) The sample preparation chamber attached to the sample stage of the light microscope. The sample container can be detached from the humidity chamber and attached to a freezing device while maintaining humidity (Takayama & Nakasako, 2012BB43). (c) LM observation of a frozen-hydrated sample of chloroplasts isolated from C. merolae, taken at a sample temperature of 82 K. The sample was frozen on a silicon nitride membrane under ∼100% RH as in §2[link] and panel (a). The bar represents 50 µm.
