Structurally variable regions and a proposed interaction interface of the cytokine receptors. (a) Cartoon representation of the structural superposition of the D1 and D2 domains of the nine receptor structures. The backbone of the selected residues (listed in Supplementary Table S2) was superimposed independently for each domain and was then drawn on top of the IFNγR2 structure. The aligned cores of both domains overlap tightly (for r.m.s.d. values, see Fig. 3[link]), but two regions in D1 (red and yellow) and one in D2 (green) are highly variable. (b) Sequences of IFNγR2 from 90 species with the sequentially most variable regions coloured cyan and conserved regions in purple drawn on the surface of the IFNγR2 structure by ConSurf (Glaser et al., 2003BB11). The IFNγR2 free cysteine Cys174 is highlighted in yellow. (a) and (b) show IFNγR2 in the same orientation.
