Unfolding of the Src and Abl chimeras in the presence of GdnHCl at 25°C in 50 mM sodium phosphate pH 7.0. The protein concentration was 2 µM, and the native fraction is plotted versus GdnHCl concentration. The samples were excited at 280 nm, and the emission spectra were collected between 300 and 500 nm. The bandwidth for slits was 5 nm for excitation and emission, and the path length was 1 cm. The centre of mass was calculated for each spectrum, and the data were normalized to facilitate comparison. |
