Key upstream steps include construct design and cloning, followed by high-throughput expression screening across multiple host systems and purification development using size-exclusion chromatography (SEC), multi-angle light scattering (MALS), dynamic light scattering (DLS), ligand-binding and activity assays. Downstream activities encompass sample preparation for structural studies, crystallization screening, cryo-EM grid preparation, data collection, and computational analysis and refinement.
