Purification of human MnSOD. SDS–PAGE of perdeuterated human MnSOD during purification. Purification steps are sequential from left to right. Lane 1, molecular-mass markers (labelled in kDa). Lane 2, whole-cell lysate. Lane 3, post-heat (60°C). Lane 4, post-anion exchange. Lane 5, post-cation exchange. Heat was applied by placing samples in a water bath, anion exchange was conducted using DE52 resin and eluting with 100 mM potassium phosphate pH 7.8, and cation exchange was performed using a CM column and eluting with a sodium chloride gradient.
