Crystals grown by the conventional vapor-diffusion method (a) (lysozyme) and by the modified vapor-controlling-diffusion method (hanging drop) (b, c, d). Crystals in the modified vapor-controlling set-up with a barrier (hole diameter of 0.3 mm) are lysozyme (b), trichosanthin (c) and BmK Mu9 (d). Photographs (e) to (h) show the number and size of the lysozyme crystals grown in hanging-drop mode using different barriers with distinct hole diameters of 0.8 mm (e), 0.3 mm (f), 0.1 mm (g) and 0.05 mm (h). These photographs were taken at the same magnification after 13 days of experiments. Compared with the case of no barrier (a), the results shown in (e)–(h) indicate that the number and size of the crystals depend on the hole diameter of the barrier. All photographs are taken over a complete hanging drop using a CCD system.
