Workflow for the CD crystal optimization for TTR. Crystallization screens were first conducted in small-diameter capillaries (top box, green border), including X-ray diffraction analyses. Work then moved to large capillaries. Here 1 mm capillaries were used as well as agarose gel mixing methods to alter crystal growth behaviour (see Section 3.4[link]). Further X-ray diffraction studies were used to check data quality and crystallization behaviour in larger-volume crystals. Preliminary neutron studies were also used to qualify the suitability of this crystallization method. Further details can be found in Section 2.2[link] and Table S1.
