Different Sx constructs and experimental setups lead to different functional outcomes for Munc18-mediated SNARE-complex formation and membrane fusion. Munc18 has been observed to inhibit SNARE-complex formation when the cytosolic domain of Sx was immobilized to affinity beads by the N-terminus, or when the N-terminus is trimmed inadvertently by protease treatment, or when the Sx is free in solution. (b) Schematic representation of in vitro experimental designs where the Sx constructs were immobilized (affinity beads) or anchored (liposomes/TMD) at their C-terminus. In these experimental setups, SNARE-complex formation was not inhibited in the presence of Munc18. See Table 1[link] for more details.
