Light and X-ray microscopic images of a wet fixed COS-7 cell. COS-7 cells were fixed with 2.5% glutaraldehyde, embedded in a thin layer of 0.1 M cacodylate buffer at pH 7.4 and observed with an X-ray microscope. (a) Differential interference contrast microscopic image of a cell; (b) X-ray microscopic image of the cell. The figure was obtained by combining several X-ray micrographs. Localization of the X-ray dense granules (arrows) corresponds well to that of the granules under a light microscope. An arrowhead shows a mitochondrion. N: nucleus, Nu; nucleolus; B: border of the cell. Bar: 5 µm.
