While in the millimolar and lower Kd range small concentrations of ligand suffice to achieve reasonable binding-site occupancy (between 70 and 90%), quite impractical concentrations of ligand in the crystallization drop are required for poor binders. On the other hand, given a sufficiently high concentration, even weakly binding (and non-native) ligands can be forced into a binding site. Figure from Pozharski et al.
