Schematic diagram of the protocol of the large-scale hanging-drop method consisting of the following six steps: (a) The seed crystals are obtained by the conventional hanging-drop vapor-diffusion method. (b) Crystals smaller than the seed crystals are removed from the droplet by nylon loops. (c) A cut pipette tip with grease at one end is attached to the glass slip in such a way as to surround the droplet. (d) The protein–precipitant mixture is injected into the cut tip. (e) The glass slips are inverted to seal onto the well plate. (f) Larger crystals are grown.
