Depolarized oblique illumination dark-field microscopy (DOIDM) of a supersaturated glucose isomerase solution (90 mg ml−1 in 100 mM HEPES buffer pH 7.0 with 200 mM MgCl2 and 1 M ammonium sulfate). The two images were taken ∼40 min after the onset of the crystallization process in (a) parallel and (b) crossed polarizers. The time shown in the images is measured after the beginning of video recording. Judging from their changing shapes the intensity peaks in (a) are clusters. The cluster indicated by a red arrow in (a) is not seen in (b). The three intensity peaks in the bottom right corner of (a) are also seen in (b), suggesting that they represent small crystals within clusters. The time interval between the images is owing to the adjustment of the analyzer.
