The spacings between the surface-exposed histidines are several nanometres and are comparable to the loading density of His-tagged proteins bound to Ni–NTA beads (Hayworth & Hermanson, 2014BB6), which presumably allows multiple binding sites to Ni–NTA and contributes to the observed affinity of Rho for Ni–NTA resin.
