Enzymatic activity of native active-site mutants of FtFBPase. (a) The enzymatic activity of native FtFBPaseII and its T89S and T89A mutants versus F16BP (substrate) concentration. The T89S mutant has a lower Vmax than the wild type by approximately fourfold (Table 3[link]), while the T89A mutant is inactive. (b) Plot of the ratio of the rates for the wild type and T89S mutant (VWT/VT89S) as a function of substrate concentration (F16BP). This type of analysis has been suggested (Eisenthal et al., 2007BB5) to compare the activity of two enzyme variants (wild type and mutants) catalyzing the same reaction in more detail. The substrate concentration has to reach a level above the Km for both enzymes to show a constant ratio between the two enzyme rates.
