The natural initiating methionine residue of wild-type TrpR is designated 1. Numbering of N-terminal extension residues begins with Pro -1 adjacent on the left to Met1, and continues with negative numbers to the new initiating methionine residue preceding the His6 sequence. Protease cleavage on the C-terminal side of Gln -3 yields the cleaved protein with additional N-terminal residues Gly-Pro preceding the natural methionine residue.
