The plots of the difference in fluorescence versus temperature were obtained using a real-time PCR system based on four replicate reactions with and without ligands. The median derivative T_m values are indicated as black dotted vertical lines. A final concentration of 0.15 mg ml⁻¹ MtRv1421-NTD was mixed with a ligand (each at 5 mM) along with the Protein Thermal Shift dye.
