An illustration of experimental optimization by tuning aluminium attenuator thickness on the sample to vary X-ray flux on the sample (all the experiments were conducted at 400 mA NSLS-II ring current). The plots show Alexa488 hydroxyl radical dose-response rates obtained for 4.0 µM Alexa488 in PBS pH 7.4 buffer for various attenuations as a function of various combinations of (a) 0.3 mg ml−1 of lysozyme and 80 µM of GDP, and (b) 0.3 mg ml−1 of lysozyme and 160 µM of HEPES.
