The squares within the grid indicate the fields of data collection limited by a 20 µm × 20 µm aperture during mapping. White-colored squares correspond to data collection at the fluorescent labels shown in (b). The aperture size in this case was limited to 20 µm × 20 µm. For each data point, a spectrum was acquired using 64 co-additions and Happ-Genzel apodization at 4 cm−1 resolution. A background spectrum through air (no sample) was collected for absorbance reference. (b) Fluorescent image of the same field. Calcein (green) and alizerin complexone (red) indicate bone formed one and two years, respectively, after ovariectomy.
